First, EMT is the conversion of malignant epithelial cells into cells with a mesenchymal phenotype (Larue and Bellacosa, 2005).
First, EMT induction endows normal and transformed mammary epithelial cells with stem cell properties, including the ability to self-renew and efficiently initiate tumors [ 106].
A recent review by Becker et al. has summarized the current state of knowledge with regard to the E-cadherin repressor SNAI1, the first EMT inducer originally implicated with tumor progression [ 9].
To our knowledge, however, this is the first EMT line of HNSCC with resistance to EGFR-targeting agent and would thus offer a useful in vitro model to understand the mechanism underlying the link between EMT and gefitinib resistance.
In this model, Snail1 and ZEB2 play a role in inducing the first EMT steps that lead to the initiation of the invasive process, whereas Snail2, E47 and ZEB1 favour the maintenance of the migratory, invasive phenotype, and Twist1 has a critical role in the development of distant metastases by prompting cancer cells to enter the bloodstream [ 12].
