Recent advancements in extraction methods should also be attempted if initial extraction success is low [80].
In total 202 chimpanzee samples yielded usable genotypes, resulting in a 46% (202/444) extraction success rate over the four year period.
Extraction success was not obviously related to time of year (data not shown) and was consistently low every year, ranging from 41% (in 2007) to 63% (in 2005).
We recommend conducting a pilot study to evaluate sample extraction success, since it can be a major limiting factor as evidenced in this study.
We show in this study that despite reliance on opportunistically collected faecal samples with poor extraction success (46% on average), we can obtain useful population estimates, albeit with moderate 95% confidence intervals.
Knowing that samples collected in successive years can be combined to obtain a population estimate is encouraging, as no estimate could be calculated for three of the four study years because an insufficient number of samples were available due to the poor extraction success rate.

After data extraction, hybridization success for each probe was inferred if flag "glsFound" values was equal to 1 (see Methods).
All patients undergoing device extractions are prospectively entered into a computer database recording patient demographics, comorbidities, device and lead type, reason for extraction, procedural success, and complications.
Many factors may contribute to this effect including the quality of the sequence assembly, degree of divergence between the organism or organisms used to extract DNA, success of extraction and labeling of high molecular weight DNA, genome size and genome complexity.

DNA extraction and sequencing success varied with the source of the tissue, the mode of preservation, and with the species group, but we did not track success rates due to changes in analytical protocols over the four years of this study.
