The purpose of this in vitro study was to investigate the amount of quadriceps force required to extend the knee during an isokinetic extension cycle before and after total knee arthroplasty.
The cycling conditions were 1 cycle 94 °C for 30 s, followed by 36 cycles of 94 °C for 30 s, 55 °C for 30 s, 72° for 45 s, and a final extension cycle of 7 minutes at 72 °C.
Enantiomerically pure polyhydroxylated natural products are synthesized by using a reiterative two-carbon extension cycle consisting of four steps.
The amplification reactions occurred at 94°C for 5 min, followed by 30 cycles (94°C 30 s, 62°C 30 s and 72°C 1 min) and finally an extension cycle of 10 min at 72°C.

The second step of the hyphal extension cycle is an EED1- dependent expression of UME6 (Fig. 9).
This may explain why mutant cells lacking key elements of the hyphal extension cycle lose their ability to maintain polarized hyphal growth and therefore switch from germ tubes to yeast cell growth.
This PCR was carried out in a thermocycler GenAmp PCR System 9700 (AB) with one cycle of 95°C for 15 minutes and then 35 cycles of 94°C for 30 seconds, 58°C for 90 seconds and 72°C for 90 seconds with a full extension cycle of 72°C for 10 minutes.
In brief, 50 ng of DNA was used in the reaction; PCR program started with 94°C for 5 min, 35 cycles of 94°C, 57°C and 72°C, a final extension cycle of 72°C for 6 minutes and stored at 4°C.
The amplification protocol for the first- round PCR was one cycle at 94°C for 2 minutes, followed by 30 cycles at 94°C for 10 seconds, 55°C for 30 seconds and 68°C for 8 minutes, ending with a single extension cycle at 68°C for 10 minutes.
Fluorescence was measured at the end of each extension cycle in the FAM-490 channel.
