Here, the following experiment is performed: (a) change the relative distance between the AFP and the CG for different depths in the rat brain; (b) calculate theoretically the expected aberrations, which are depth-independent (see Appendix A); (c) measure the aberrations experimentally with CGWS and compare them to the theoretical predictions.
Our data permits estimating the degradation of image quality with depth as well as the possible improvement that can be expected from aberration correction.
As it was expected the aberration effects remained moderate while scattering effects resulted in the requirement of an efficient excitation strategy to image the sample.
For the 20×/0.5 objective, the expected spherical aberration slope is too small to be accurately measured by CGWS (it corresponds to a RMS wavefront error of λ/162 when the CG is displaced by 10 µm).
In turn, this permits estimating not only the gain in signal and resolution that can be expected from aberration correction, but also the distortions that cannot be corrected using adaptive optics.
However, since the same limitation applies to the correction of the aberrations, the measured part corresponds to the part that can be corrected: consequently, although the uncorrected residual wavefront cannot be measured, the gain that can be expected from aberration correction can be accurately determined.
As expected, many aberrations are common, but interestingly, some aberrations are present in early biopsies and not in later ones.
With such fluences, the number of cells with induced aberrations per nucleus irradiated increased up to 4-fold in CHO cells and 15-fold in xrs-5 cells over that expected if aberrations occurred only in irradiated cells.
As expected, genomic aberrations were detected by 44K array CGH as previously detected by our custom-made BAC array CGH in all of the 13 PTCL, NOS samples analyzed (Fig. S1).
As expected, most aberrations observed in B-LCL cell lines were also observed in all CD19+ cells: deletions at the Ig heavy chain on chromosome 14 and at the Ig κ and λ light chains on chromosomes 2 and 22; amplifications at the TCR-α locus on chromosome 14, and at the TCR-β and γ-loci on chromosome 7.
Trademark von Trier, one might say, harping on a weird waif's woes: but made idiotically pretentious by dim, tuneless, ineptly staged song-and-dance interludes, supposedly the girl's fantasies, and allegedly inspired by Jacques Demy's "Les demoiselles de Rochefort".After this long-expected aberration, the jury, like the festival, went east.
