In some cases the observed experimental heterogeneity can be attributable to pre-existent variant cells within tumor cell populations [ 18, 24], but even within groups of syngeneic animals treated with the same tumor cells, the numbers of resultant metastases can be quite variable [ 25, 26].
This is a somewhat conservative threshold as the probability that a non-existent variant is detected three times or more at the discussed coverage depth is very small at an overall sequencing error rate of 0.5 1% if the errors were to occur independent of each other.
