The estimation problem can be regarded as equivalent to the accurate reconstruction of ionic current kinetics and amplitudes in an excitable cell model, given only action potential experimental data.
On one hand, it is plausible that in an excitable neuron model, in which the crossing of a threshold may be aided or prevented by a weak driving, addition and deletion of spikes as in our AD model can be observed.
Excitable cell modelling is of a prime interest in predicting and targeting neural activity.
To further investigate this issue, we built a probabilistic excitable cellular automaton model [11] tuned near the critical state (Fig. 4a), and deliberately undersampled it with a spatial structure equivalent to that of our MEAs (Fig. 4a, inset; see also Methods).

Interspecies differences can limit the translational value of excitable cells isolated from model organisms.
The canonical model for excitable media are the well-known FitzHugh Nagumo equations [26] with diffusion in the activator variable: ε ∂ u ∂ t = u − 1 3 u 3 − v + ∇ 2 u, ∂ v ∂ t = u + β. (1).
Note that we work in a regime where the model is excitable, as we do not include any background drive that would be able to make the single neuron model fire in the absence of synaptic coupling or head-direction input.
The observations are consistent with a model with excitable dynamics where the first change is rapid and noise-triggered followed by slow relaxation to the initial state [3].
