Immunohistochemical staining of a lung autopsy has been performed by using a monoclonal antibody directed against the hMPV matrix protein and revealed evident localization of the M protein in the cytoplasm of bronchiolar epithelial cells.
The R121A mutant retains some function compared to a cdc8-27, a temperature-sensitive mutant that is unable to initiate the first stages of contractile ring assembly at the restrictive temperature: Rlc1p-mCherry and Fim1p-mEGFP have no evident localization at the restrictive temperature (Fig. 2C,E) and Cdc12p-3xGFP iseenen as a single dot (Fig. 2D).
Three types of rDNA constellations were evident, including: localization on the autosomes (one autosomal pair) in about half of the species; localization on the heterosomes (one of the X chromosomes and the Y) found in six species; and localization on both (one autosomal pair plus heterosomal copies located on one of the X chromosome) in the remaining five species.
During necropsy, multiple, well-defined, whitish nodules were observed in most muscular and subcutaneous tissues, with no evident preferential localization.
In positive cell lines, endogenous DAAM1 showed evident co-localization with actin stress fibers particularly in the sub-nuclear region (Fig. 1C, ventral section), and on centrosomes (white arrowhead in Fig. 1C medial section).
In contrast, in the presence of either myc-K110ADMPK or myc-ΔMADMPK mutants, YFP-InsR was retained in intracellular structures (Fig. 5 A, i l and m p, respectively), with no evident co-localization with the DMPK mutants (Fig. 5 A, k and o).
The confocal microscopy technique, applied in order to identify this cellular type, demonstrated an evident co-localization between PrPsc accumulation and astrocytes.
The relationship between PrPsc and glial cells observed by conventional immunohistochemistry, was confirmed by confocal microscopy showing an evident co-localization.
As presented in Fig. 5, no evident co-localization of EpoR and Hep Par-1 was detectable by immunostaining, indicating a very low expression of the receptor in rat hepatocytes (Fig. 5a, c).
Additionally, ER-α has an apparent intracytoplasmic localization without any evident membrane location (Sirianni et al. 2008).
An evident nucleolar HEX localization was observed also in T47D cell line; in this cell line the cytoplasmic expression of HEX was more evident than that observed in MCF-7 cells (Fig. 1, panel F).
