GCM can provide an estimate of connection directionality when one time-series resembles a time-shifted version of the other, supposing that one with temporal precedence caused the other [ 20]).
Instead, we calculated point estimates of the connection coefficients using the TLSR method [ 16].
Repeat injections enable identification of a few additional sparse projections and also provide better estimates of average connection weights.

If corresponding treatment effect estimates of various connections, or so called paths, differ between two treatments, there is inconsistency.
Specifically, a single-injection approach can 1) detect all but the most sparse projections with high probability, 2) provide a reasonable estimate of the connection weight of each pathway (generally within an order of magnitude), and 3) identify some of the sparse connections that are statistically likely to be inconsistent across multiple injections.

Despite this qualification, our results suggest that techniques with intermediate temporal resolution may yield good estimates of the functional connections between individual neurons.
