Furthermore, despite some rare ESC treatment failures verified in recent years [ 3– 14], the anticipated number of reported gonorrhoea treatment failures has not yet materialised, which could also be partly due to a lack of identification and under-reporting of treatment failures.
Most of these ESC treatment failures have been caused by MDR N. gonorrhoeae strains belonging to the MLST ST7363 and ST1901 [ 3, 4, 7, 9, 12, 13].

However, worryingly the proportion of isolates displaying decreased susceptibility to ESCs (MIC = 0.125 mg/L), which have previously resulted in gonorrhoea ESC treatment failures [ 13, 15, 16, 44], rapidly increased from none in 2010, to 7.6% in 2011, and 13.9% in 2012.
The revised 2012 European Society of Cardiology (ESC) chronic heart failure guideline was used to provide a contemporary definition of heart failure.
Inducible knockout of PDCD2 in ESCs resulted in failure of entry into S phase of the cell cycle, and PDCD2 knockout embryos exhibited a significant defect in growth.
One isolate from India also belonged to ST1407, which has been shown to be a internationally spread successful gonococcal clone that accounts for the majority of the decreased susceptibility and resistance to ESCs and treatment failures with cefixime worldwide [ 12, 17- 19, 61, 62].
The low rate of chimera formation may be caused by a problem of the ESCs to integrate into the inner cell mass, and/or from failure of the incorporated ESCs to truly participate in embryo development.
Taken together, analyses of gene expression at the single-cell level indicate that the increase in steady state mRNA levels detected by quantitative RT-PCR in Mbd3 −/− ESCs is due to the failure to restrain transcription levels of actively transcribed genes irrespective of culture conditions, as well as a failure of gene silencing in a subpopulation of cells grown in serum and LIF conditions.
The degree of Stat3 activation by extrinsic factors might be subtly different among ESCs from different species, and these subtle differences might account for the failure to establish authentic ESCs from non-rodent species under the LIF condition.
As shown above, DNA content analysis of both MEFs and ESCs depleted of PDCD2 revealed a failure to enter S phase; however, neither MEFs (Fig. 1B) nor ESCs (Fig. 2B; supplementary material Fig. S6A), showed increased cells containing sub-G1 amounts of DNA, suggesting that PDCD2 loss did not lead to apoptosis in these cells.
