This integrated approach to reforestation of ROK was implemented in three pillars of action: 1) coordinated national plans, 2) collaboration among the governmental branches, and 3) organizational reformation.

To determine if CagA indeed requireed Rho and Rok to redistribute MLC, we used RNA interference (RNAi) to knock down expression of Rho and Rok, as well as a chemical inhibitor of Rok, Y-27632, to determine if CagA was able to redistribute MLC-GFP even in cells depleted of Rho and Rok activity.
Interestingly, we have shown that the essential role of Rok in cytokinesis is simply to maintain phosphorylation of myosin II RLC, as shown by the rescue of all Rok depletion cytokinesis phenotypes by expression of phospho-mimic RLC.
In contrast, reducing levels of Rok via rok RNAi in RhoGEF2 + Raf GOF clones rescued the tumorigenic phenotype (Fig. 3).
Given the finding that Rok suppressed RhoGEF2 + Raf GOF cooperation, we then sought to examine which effectors downstream of Rok are required.
Alternatively, tumour cells might accumulate mutations, downstream of Rok- α or in other pathways, which enable them to tolerate high Rok- α activity without undergoing differentiation.
