Objective: To validate a new combination of a viability assay and Fluorescence in Situ Hybridsation (FiSH) techniques using X- or Y-DNA probes to assay the viability of each single population within bidirectional mixed lymphocyte cultures (MLCs).
The differentiation of each single population by FiSH showed, as in the metaphase assay, that the relationship between viable maternal and neonatal cells was shifted significantly (p<0.05) towards the neonatal cells, while the relationship of the viable cells in the MLCs between unrelated adults was balanced.
Each single population data set was initially filtered to remove SNPs and individuals with a call rate below 0.9.
Since BRCA2-8765delAG occurs within a triplet repeat sequence of AGAGAG, our study further confirmed the existence of a mutational hot-spot at this genomic position (additional genetic factors within each single population might be involved in generating such a mutation).

The hybrid line was created with a four-generation crossing design (Figure S2), that, in the first three generations, created eight lines that each had a single population's cytoplasmic component combined with all eight nuclear genomes.
