The observation that non-canonical splicing events can reproducibly occur between short repeated sequences lead us to question whether many recently reported trans-splicing events are in fact due to template switching artifacts.
Errors in DNA replication due to template slippage or unequal crossing-over can result in tandem duplication (TD), producing tandem arrays (TAR) of paralog genes in close genomic proximity [ 52].
This combination of ultraclean consumables and reagents, along with improvements incorporated into iMDA provides the opportunity for specific detection and genomic sequence analysis from samples that were previous not possible due to template limitation.
Ladoukakis and Eyre-Walker reported a small but significant excess of short inverted repeats (6 9 bp) in protein-coding sequences and proffered that the inverted repeats could arise by sequence-directed mutagenesis where an imperfect palindrome or inverted repeat converts to a perfect one due to template switching during replication.
From these data, it is tempting to speculate that the five Sμ mutations accumulated during disease progression from an Sμ-unmutated ancestor clone, however, as the Sμ primers, unlike the IgV primers, can also specifically bind to DNA from non-B cells, the analysis of Sμ mutations likely assumes a higher risk of PCR artifacts due to template contamination with DNA from non-B cells.
Due to template effect of different guest cations, 1 presents an interesting 2D smectite-like lamellar framework that formed by the 4-linked (4,4) anionic zinc terephthalate polymeric network and the interlayer [Ph3PCH2Ph]+ exchangeable cations, while 2 shows a 2D 3-linked (6,3) H-bonded anionic zinc terephthalate polymeric brickwall network with encapsulated guest [Ph4P]+ cations.

The frameshift mutations suggest that replication becomes unstable due to template-switching errors.
Figure 4 shows 62.4% of reads with Q20<100 bp fail as a result of process-related problems while 75.5% of reads in the Q20: 500–599 bp bin fail due to template-related characteristics.
In this case, the use of the thioanalogue instead of citrate in a specific reaction step is not due to template-copying (matching) but due to the fluctuations in external factors, such as the availability of resources, nature of the environment, temperature and so.
It is important to be cautious with the interpretation of "non-conventional" RT-PCR products such as chimeric transcripts since their existence can sometimes be attributed to experimental artifacts due to template-switching activities of the reverse transcriptase (RT) or Taq polymerase [ 16].
