Simultaneously, OBs were seeded onto 6-well culture dish inserts (BD Bioscience, USA) at a density of 3 × 105 cells per insert with 2 ml of media.
The first trimester (5 8 weeks) placental villi were dissected into the explants of 2 5 mm in diameter, and explanted in Millicell-CM culture dish inserts (0.4 mm pore size, Millipore, Carrigtwohill, Co., Cork, Ireland).
Briefly, small fragments of villous tips (15 20 mg wet weight) were placed on Millicell CM culture dish inserts (Millipore Corp., Bedford, MA, USA) previously coated with 180 μL undiluted Matrigel (Collaborative Research, Inc., Bedford, MA, USA) and then inserted in 24-well plates and left overnight at 37°C in a humidified atmosphere of 95%air/5%% CO2 to allow explant attachment to the Matrigel.

In brief, small pieces of tissues (2 3 mm) from tips of first trimester human placental villi (5 w–8 wereere dissected and explanted in Millicell-CM culture dish inserts (0.4 μm pore size, Millipore, Carrigtwohill, Co., Cork, Ireland) pre-coated with phenol red-free matrigel substrate (Becton Dickinson, Bedford, MA, USA).
Cells were washed with PBS, the media replaced, and the dish inserted in a closed, thermocontrolled (37 °C) stage top incubator (Tokai Hit Co., Shizuoka-ken, Japan) atop the motorized stage of an inverted Nikon TiE fluorescent microscope (Nikon Inc., Melville, NY, USA) equipped with a 60× oil immersion optic (Nikon, CFI PlanFluor, NA 1.43) and NIS Elements Software.
The polyethylene insert is manufactured from ultra-high molecular weight polyethylene (UHMWPE) (GUR1020) by direct compression molding from powder, which produces sufficient durability [ 9] with deep-dish insert shapes.
Cultures were carried on culture dishes inserts in the presence of the inactivated with Mitomycin C monolayer of 3T3 fibroblasts.
As described here, smart Pétri culture dishes inserting a PRP allow actual use of the technique in live cell experiments.
