The highly de-differentiated passage 5 (P5) chondrocytes failed to respond to the stimulation by either growth factor.
ATDC5 cells [ 9] (3 × 104 cells/35 mm dish) were maintained in DMEM/Ham's F-12 (1:1) containing 5% fetal bovine serum, 2 mM L-glutamine, 10 μg/ml human transferrin, and 3 × 10-8 M sodium selenite (maintenance medium), or the cells were differentiated, without passage, in the same medium supplemented with 10 μg/ml insulin (chondrogenic medium).
