Process variability in RNA isolation, preparation and analysis was investigated (data not shown), confirming that the majority of the observed variability between replicates does indeed represent biological differences between cultivations.
This regulatory impact differs between cultivation in light or darkness.
Sustained, efficient RFP expression was noted in rAAV-RFP-transduced aspirates both in dynamic and static culture conditions already after 7 days and for at least 28 days (the longest time point examined) (Madry et al. 2013) compared with control transduction (absence of vector treatment), showing no visible differences between the two cultivation systems (Fig. 1b).
