H4K20me3 is not enriched to a greater degree at developmentally repressed imprinted genes compared to expressed genes which again is inconsistent with a role in developmental repression; in fact a higher incidence is observed at developmentally expressed compared to repressed genes (Additional file 2).

Our findings suggest that the germline mark crucial for the establishment of genomic imprinting is specifically acted upon, at least in ESCs, by different repressive epigenetic mechanisms than those regulating more canonical developmental repression.

This implies that this particular histone modification profile is associated with developmental repression and, therefore, that these two modifications often mark the developmentally regulated, 'normally active' allele when repressed.

Furthermore, when assessing only imprinted genes that do not have a promoter DMR and are developmentally expressed in ESCs (in order to exclude H3K27me3 enrichment involved in developmental repression), H3K27me3 is enriched only 20% of the time (n = 10; data not shown); H3K27me3 enrichment is found at 30% of developmentally expressed imprinted genes that do possess a promoter DMR (n = 10).

The results depicted in Additional file 2 suggest a propensity for H3K27me3 to play a role in developmental repression of imprinted genes: in mouse ESCs H3K27me3 is enriched to a greater degree at developmentally repressed imprinted genes (53%) than at expressed genes (25%).

Additionally, H3K27me3 is associated with developmental repression of imprinted genes as discussed above.

Despite the fact that VELVET has initially been reported for its involvement in red-light initiated conidiation in A. nidulans [ 135, 136], recently functions exceeding developmental regulation, namely repression of penicillin biosynthesis in this fungus [ 137] have been shown.

The BOB1 gene, in cooperation with the AS2 (AS1) gene, plays a crucial developmental role via repression of both the indeterminate state and the abaxial fate of cells in the presumptive adaxial domain of leaf primordia after the commitment to leaf initiation around the shoot apical meristem, promoting the establishment of the adaxial polarity of leaves (Fig. 8).

A key component implicated in the establishment of this epigenetic signature in the regulation of ESC is the Polycomb group family of proteins, which are responsible for maintaining the pluripotent state by epigenetic repression of developmental genes through the presence of repressive chromatin marks in the promoter regions of genes [10].

And although the regime seeks to appeal to sectarian divisions and invoke the threat of Iran in order to delegitimise dissent, the truth is that the discontent is found across Saudi society, fed by political repression and developmental failure, as a result of corruption, government malfunctioning, and the squandering of billions on arms.

Through epigenetic mechanisms, such as site-specific targeting and higher-order chromatin remodeling, REST and CoREST can mediate cell type- and developmental stage-specific gene repression, gene activation, and long-term gene silencing for protein-coding genes and for several classes of ncRNAs (e.g. microRNAs [miRNAs] and long ncRNAs).
