The present study demonstrated that a unique structural feature of the dependency of amyloid-precursor epitope on length of polyQ expansions and its link to nucleation are the critical determinants of the repeat-length related age-of-onset of HD.
While the main point of this study was to evaluate the wisdom of focusing solely on repeat hits in postscreening efforts, the analysis of the determinants of binding of the repeat hits to the anti-ADP3 antibodies was also carried out and provided interesting data.
In particular, two STRs, (TAGAA) j and (TAAAA TAGAA) m, form ∼90% of the entire repeat expansion, and the values of j and m are positively correlated (correlation coefficient r = 0.70), implying that these two values are the determinants of the instability of the repeat expansions in SCA31 (Fig. 4D).
