When considering phenol chloroform based extraction methods (BP1 and BP2 methods) the DNA quantification and purity determination using UV absorbance at 260 nm may be error prone due to the high extinction coefficient of phenol at 260 nm resulting in lower detectable yields (Yang 2008).
