Cognitive psychology studies have described pooling in illusory conjunction, which is the false perception of an object that is not present yet is composed of "features" from objects that are present (Prinzmetal, 1995; Robertson, 2003; Treisman, 1996; Treisman & Schmidt, 1982).
While there are a variety of reports describing pooled sequencing or multiplexed solution hybridization capture [ 10- 18], our method offers a highly uniform, reproducible workflow for smaller labs that may not have access to high-throughput robotics but still wish to quantify rare variation within specific populations.
To determine whether any of the remaining chromodomain-containing H3K9me3 readers expressed in mESCs are required for SETDB1-mediated silencing, we knocked down Cbx3 (HP1γ) as well as Cdyl2, Cbx1, Cbx2, Cbx5, Cbx7 and Mpp8 in the above-described reporter lines and a previously described pool of mESCs harbouring the silent murine stem cell virus (MSCV) provirus [ 20].
