For a pathway P with set of genes g 1, g 2, …, g k, the overall procedure is summarized as follows: 1) For each pair of g i and g j in P, the cooperation profile t ij, competition profile d ij, redundancy profile h ij, and dependency profile l ij are calculated for disease and control groups separately.
To identify such interactions, we use the minimum mRNA expression for pairs of genes to define the dependency profile of genes g i and g j as (4) l ij k = min e i k, e j k for 1 ≤ k ≤ m These four gene interaction profiles are conceptually illustrated in Figure 1.
In Europe and North America a handful of studies have begun to document the changing disability and dependency profiles of the oldest old [ 4- 11].

As shown in Fig. 2a, native HydA1 is most active from pH 7 to pH 8 while wild-type CpI (Fig. 2b) clearly reaches its highest activity at about pH 8. Thus, we did not exclude hits based on the iron-dependency profile but noted the ratios as a possible indicator for the compounds' mechanism of action (for example, the isoquinoline diones series, Figure 2 and Table S1).
