After 24 h, the results of the cell viability test demonstrated cell viability at 101, 91, 79, 64, and 46%% for doses of 1, 10, 50, 100, and 200 μM BC, respectively (Fig. 1a).
Forty-four drugs from the library demonstrated cell viabilities higher than 70% at a concentration of 20 μM and were screened for their ability to sensitize PC3-TR cells to TRAIL under these conditions.

Thus, about 37% reduction (demonstrating cell viability percentile change from 45%to65%5%, and cell death percentile change from 55%to35%5%) was observed in 2.5 μM Doxorubicin-induced cell death, when Cas-/ (EV) cells were compared to Cas-FL cells (Additional file 1).
Likewise, Bortezomib-induced cell death was reduced by about 45% in Cas shRNA-transfected 293T cells (demonstrating cell viability percentile change from 55% to 75%, and cell death percentile change from 45%to25%5%).

The cells always responded to both the normal Ringer's (by returning of the Ca2+ influx pattern to the normal basal level) and the Ringer's 20 mM KCl (by displaying a large and transient Ca2+ influx due to cell depolarization), thus demonstrating cell viability.
Similarly, knock-down of Cas by shRNA reduced MG132-induced cell death by about 40% (demonstrating cell viability percentile change from 65%to80%0%) in HeLa cells.
In the present study we demonstrated good cell viability at all time points post injection with little cell death as evidenced by caspase 3 immunopositivity or the presence of apoptotic bodies.
