Reconstructed image stacks were created using XMReconstructor software (Carl Zeiss Microscopy GmbH) and the subsequent segmentation (delineation) of the structures of interest in the male and female genitalia was performed with Amira 5.4.5 (Visualization Science Group, FEI).
At the genome sequence level, the International Wheat Genome Sequencing Consortium IWGSCC, http://www.wheatgenome.org/) has tackled the delineation of the genome structure using the capacity to generate bacterial artificial chromosome (BAC) libraries from flow-sorted chromosome arms followed by sequencing of BACs that were arranged into minimum tiling paths (Feuillet et al. 2012).
Adding MAR in the reconstruction process eliminated the majority of intra- and extracorporal metal- and beam hardening artifacts, resulting in a better delineation of the neighboring structures and the jaw itself (Fig. 2b).
These results provide an indication of the added-value of this sequence resource in the framework of the delineation of gene structure, especially with respect to the determination of the transcription initiation site.
This work demonstrates the utility of genome scans for adaptive divergence in the delineation of stock structure, the traceability of individuals and products, and ultimately a role for population genomics in fisheries conservation.
