No detectable mRNA expression of LH, FSH, TSH, ACTH, NeuroD1 (Oyama et al, 2001), Tpit, SF-1 (Barnhart and Mellon, 1994), and PROP1 was found in either PASC1 or the daughter cells (data not shown).
There was no significant difference in the expression levels of multiple drug resistance related genes, such as BCRP1, MDR1, MRP1, and MRP3 (Donnenberg and Donnenberg, 2005; Abbott, 2006), between PASC1 and the daughter cells (data not shown).
The progeny that had divided often produced two daughter cells unequal in size (data not shown).
Of these 16 plasmids, 14 were detected as colocalized pairs (seven signals) in G2, 12 partitioned equally to daughter cells; and two did not (data not shown).
After anaphase and cytokinesis, both daughter nuclei move toward the basal side (data not shown).
However, we could not reliably detect expression of SYS-1 in the α or β cells using the reporter that revealed differences in the Z1 and Z4 daughters, qIs95 [ sys-1p::VENUS-SYS-1] (data not shown).
Consistent with expectations, medicated hypertension was associated with known risk factors including daughters' race/ethnicity, menopausal status, BMI, diabetes, and age (data not shown), but only race/ethnicity, menopausal status, BMI, and diabetes were retained in the final model 3 based on p-values < 0.05, and associations based on this model were similar to those from model 2.
In four of these lineages, we could observe that the two daughter cells divided again during the observation period (data not shown), revealing their progenitor character.
Although we were unable to internally validate self-reported BMI in the present study, comparisons of maternal and daughter adult weight, height, and BMI showed expected increases among daughters compared to the mothers' generation for all three measures (data not shown), similar to secular trends observed nationwide (Nooyens et al. 2009; Reither et al. 2009).
Efficient formation of daughter spheroids was observed for both ALDH+ and ALDH− groups (data not shown).
