The extraction was repeated twice and the total organic solution, collected from each step of extraction, was evaporated using a rotary evaporator (Büchi Rotavapor R-114, Switzerland) that yielded the crude (M : DCM) extract.
We therefore tested the ability of a panel of E2s to collaborate with Huwe1 in mediating TBP ubiquitination, and found that both the UbcH5 family E2s and UbcH7 are active in the in vitro ubiquitination assay with recombinant purified Huwe1, consistent with the E2 enzymes that worked with the crude 0.3 M KCl fraction from the D52 column.
