To corroborate the interaction between TDP-43 and these miRNAs suggested by the raw iCLIP data, we assessed their interaction with an RNA immunoprecipitation (RIP) assay, which is similar to iCLIP, but without a UV cross-linking step (Peritz et al, 2006).
Although these findings corroborate the interaction of I αI and the complement system, several questions remain.
The results corroborate the interactions of the tested components, as the close proximity of component pairs were visualized as clear enrichment of signals on the X chromosome (Fig. 2, Suppl. Fig. 1).
Although further studies would be needed to corroborate the interactions found, our methodological approach has demonstrated to be a promising complementary tool for finding new susceptibility genes in these thyroid carcinomas.
We corroborated the interaction of hERK1 with H2A and H4 by pulldown of GST-hERK1 and immuno-staining of histones (Fig. 4D) and observed a FRET efficiency of almost 0.2 and 0.3 between hERK1-GFP and H4 or H2A, respectively, in the nuclei of HeLa cells, with a somewhat lower in the cytoplasm (Fig. 4C).
This approach corroborated the interaction of ORCA with G9a.
Specifically, ITC binding experiments with blocked SNARE complexes, where a 2.5 3-fold excess of blocking peptide was present, have corroborated the interaction of CPXacc with the t-SNARE groove in a pre-fusion (truncated) SNARE complex as observed in the crystal structure.
We further corroborated the interactions of Tfg, IKKγ and Carma-1 by Blue Native gel electrophoresis, where Carma-1 and Tfg formed a 0.7 1 MDa complex.
Our in situ PLA results also corroborate the proposed interaction between MLE and TopoII (Fig. 6).
To corroborate the physical interaction between HOT1 and Coilin, we performed IF stainings for HOT1 and Coilin in nonsynchronized HeLa cells.
The findings support the hypothesis that HNF4α is expressed in each of these types of cells and indirectly corroborate the functional interaction of gene products in the networks reported above.
