In this study, we provide a novel strategy to correct different types of natural splicing mutations using exon specific U1 snRNAs (ExSpeU1).
The ExSpeU1s we have developed here, which bind to non-conserved intronic sequences, have two major advantages, (i) they do not interact directly with normal 5′ss and (ii) correct different types of splicing defects associated to exon skipping.
Screening of U1 snRNAs binding at non-conserved intronic sequences downstream the exon identified for each case an unique exon-specific U1 snRNA (ExSpeU1) able to correct different types of mutations.
