One possible explanation for this result is that the Ulp1(3)(C580S) two-hybrid prey construct interacts with a number of available substrates in the cell (for example, free Smt3 and other sumoylated proteins), and, as a result, this sequestration is no longer available to the Smt3 two-hybrid bait construct, thus creating the appearance of a weak interaction.
