The conformation and quantity of proteins adsorbed onto a surface depend on numerous factors, which include: surface chemistry, roughness, and local geometric characteristics such as curvature.
Changes in the type, conformation, or quantity of bonds generate different spectra; this could be used to differentiate between benign and malignant tissues.
To exclude FRET signal from Fn adsorbed onto the bottom coverglass, we discarded the slice z = 0 and used the slice z = 2 μm (above the coverglass) to quantify Fn conformation and quantity adsorbed at various HAP concentrations.
