These encouraging results show that de novo design of an idealized protein structure of more than 200 amino acid residues is now possible, from construction of a particular backbone conformation to determination of an amino acid sequence with an automated sequence selection algorithm.
For both the TRPV1 channel and ASICs, these toxins were used to stabilize specific conformations for structure determination by cryo-electron microscopy and X-ray diffraction (Baconguis and Gouaux, 2012; Dawson et al., 2012; Baconguis et al., 2014).
In this context, the determination of conformation becomes a matter of prime importance.
Using EM we find that apo- DmORC is conformationally heterogeneous, but that the non-hydrolysable nucleotide analog ATPγS locks the complex in a specific conformation that permitted the determination of its 3D structure to 22 Å resolution.

Applying the simulations to the MreB mutants I identified previously, I show that those cell-shape mutants have altered MreB filament conformations, highlighting the molecular basis of cell shape regulation and determination.
The information on PBLG molecular conformation provided by IR determinations indicates that in the acid mediated liquid crystal solution, the PBLG side chain may become flexible while the helix backbone remains stable prior to the helix coil transition.
