Here we compare simulations that have been shortened by 450 km using the rheological parameters of Supplementary Fig. 2a and Supplementary Table 1 (and the weak zone configuration shown in Fig. 2a).
One way to achieve this would be to maintain the 'channel' open for passage to the HIV-activated attractor by not switching off the activation, so that the TAT nullcline will stay in the configuration shown in Fig. 2k (left).
The maximum stress on the nanobeam is calculated to be much lower than the transverse rupture strength for the configuration shown in Fig. 1, and therefore fracture is highly unlikely.
Next we tested the second configuration shown in Fig. 2b and 3, which involves the reflective loop set-up and allows the Omnipolarizer to operate in (or switch among) the two different regimes – a polarizer or a PBS.
In the experimental configuration shown in Fig. 2, the combined use of HWP2 and PBS facilitates flexible output power adjustment by properly changing the relative angle between HWP2 and PBS.
Therefore, the fact that the intermittent dosing scenario spends time in the phase space/nullcline configuration shown in Fig. 2f during times of low drug concentration is not compensated by more cells entering the activated HIV basin of attraction during times of higher drug concentration.
By contrast a continuous dosing scenario with identical drug exposure will be fixed in the phase space/nullcline configuration shown in Fig. 2k such that the 'channel' for state switching (cells move along the HDAC nullcline from inactivated HIV attractor to activated attractor) will remain open, resulting in a higher frequency of HIV reactivation.
