Using a high-throughput multifactorial cellular array technology, we investigated the effect of hundreds of ECMP combinations and concentrations on differentiation of several hPSC lines to definitive endoderm (DE), an early embryonic cell population fated to give rise to internal organs such as the lung, liver, pancreas, stomach, and intestine.
Increased concentrations of growth differentiation factor 15 (GDF-15) and high-sensitivity troponin T (hsTnT) were found in patients with both HFpEF and heart failure with reduced ejection fraction (HFrEF) as compared to community-based controls [ 50].
In a previous study we showed that MPEs prepared with n-octyl-glycoside induce a concentration-dependent inhibition of differentiation in OPCs cultured in differentiation medium for 48 h similar to that observed with crude myelin membrane preparations (Syed et al., 2008).
Consistent with this hypothesis, we found that the endogenous levels of RASSF9 mRNA were initially induced when keratinocytes were exposed to a calcium concentration of 2 mM (differentiation-inducing conditions) for 1 day, but were thereafter reduced as differentiation progressed (Figure S10).
To investigate the effects of pre- and postmenopausal serum with their inherent hormone status on steroid hormone signaling, myotubes at day five were exposed to the serum at final concentration of 10% v/v of differentiation medium.
Tau protein was detectable as a single band at low concentration from Day 30 of differentiation, and the levels of tau increased dramatically between Days 20 and 30 of differentiation, which coincides with the appearance of post-mitotic neurons during differentiation (Fig. 2Bi).
