The crude product was purified by RP-HPLC using full H2O/AcN (0.025% TFA) gradient yielding colorless oil (25 mg, 74%).
After stirring overnight at ambient temperature, the solvent was evaporated under reduced pressure and crude product purified by Flash chromatography using DCM/ethyl acetate (EA) gradient giving colorless oil (108 mg, 0.18 mmol, 70%).
After drying the samplet at 50 °C for 2 h, automated chromatography using a petroleum ether/EtOAc gradient (0 40 %) gave 4-cyanobiphenyl as a colorless solid (149 mg, 83%%) with the following physical properties identical to a reference sample: M.p. 84 °C (ref. 54 84 85 °C).
D3 was purified by semi-preparative HPLC using a gradient elution (MeCN/H2O 50 50 2.5 mL/min) to give compound 1 (4.4 mg, colorless oil).
The crude product was redissolved in water, lyophilized, and subsequently purified by preparative reversed phase HPLC using an acetonitrile/water gradient containing 0.1% TFA (Merck Chromolith prep RP-18e column, 100 × 25 mm) resulting in a colorless solid after lyophilization for a yield of 56 mg (60%).
It was colorless.
Imperceptibly the gradient increased.
