Eco RI and Hind III digestion of plasmid pCR 2.1 results in the deletion of a short 65 bp fragment (Fig. 1B). Figure 2 shows the Lig-PCR products obtained by direct PCR amplification of TA and cohesive ligation reactions using M13 forward (-20) and M13-reverse primers (Fig. 1).
