Currently, the most advanced current array technology, Infinium MethylationEPIC BeadChip array, offers a unique, logistic and holistic approach, allowing assessment of the methylation status of not only 5mC but also 5hmC, including 2880 CNG sites (where N is any nucleotide and C and G are cytosine and guanine) and 59 SNP sites (Moran et al., 2016).
However, there are recent advances on the structure of the maize crystallized CMT3 protein, highlighting its domains involved in binding to nucleosomes and catalysis on CNG sites.
Methylation of CG and CWG sites is maintained by different mechanisms: CG sites are maintained by a plant homolog of mammalian Dnmt1 acting on hemi-methylated DNA after replication [ 57] whereas methylation of CNG sites (N is any nucleotide) is maintained by other mechanisms.
In contrast, very few of the numerous CNG and CNN sites were methylated under either normal or non-water stress conditions (Fig. 1B).
The first group should not be addressed to anti-EGFR MoAb treatments based on the sole primary tumour evaluation, but eventually, it may benefit from these targeted therapies because of the presence of EGFR gene CNG in metastatic sites.
Notably, the cGMP binding site of CNG ion channels have a threonine at this position, and like PKG I substitution of this residue with alanine decreases cGMP sensitivity of the channel 30-fold without changing its cAMP sensitivity [17].
This enhanced reactivity could also be due to more active sites available in CNG soot, as well as due to CNG soot being immature.
The cGMP-binding site of PKG and CNG channels has a threonine residue distinct from the cAMP receptors, and previous models based on the known structures of PKA and HCN channels have predicted that the hydroxyl group of these threonine residues interacts with the guanine 2-NH2 grofp of syn-cGMP through hydrogen bonds.
Direct actions of cGMP can be exerted by binding to agonist or regulatory sites on cyclic nucleotide-gated (CNG) ion channels (reviewed in Kaupp & Seifert, 2002) or hyperpolarization-activated, cyclic nucleotide-modulated (HCN) channels (reviewed in Craven & Zagotta, 2006).
