The authors also observed that periplocin showed an inhibition of tumor growth when human Huh-7 hepatoma cells were injected into immunodeficient mice, without observing clear side effects on the host.
"It has a very, very clear side-effect profile," Yaffe says.
This was not the case, however, because there was no clear side-effect [F 1,4) = 6.33; p = 0.065] on the duration of the monopodal stance.
The results may suggest that clear-cut side-effect profiles from premarketing RCTs are less distinct in a naturalistic setting where samples are more heterogeneous and concomitant medications less restricted.
"You need a much clearer side-effect profile and labeling to get the broad adoption you have with Plavix," Mr. Schott said.
While Fmr1 KO mice improved both their procedural memory formation and avoidance discrimination after administration of Fenobam, WT mice showed a clear negative side effect in that their procedural memory formation was severely impaired.
Grippo believes that careful dosing would help users steer clear of side effects.
"For me, it was like, if I was going to put a drug in a game, I think it was okay, because you take it, and it's clear the side effects are really, really bad," says Jorgensen.
Reasons for non-compliance are not clear but side effects are likely to be one cause.
