Conversely, the development of a circumferential collagen network was only predicted at these pressure conditions when cell contractility was high.
In absence of EGF, short actin bundles were distributed radially in the cells located at the periphery of growing colonies, whereas a well-developed circumferential actin network was observed in cells of terminal colonies (Fig 2A).
The effect of NDGA on the filamentous actin cytoskeleton was comparable to that of cytochalasin D. However, in addition to the disruption of actin stress fibres, cytochalasin D also disrupted the circumferential actin filament network.
