Human recombinant IL-4, soluble IL-15Rα/Fc Chimera (s-IL-15Rα), soluble IL-21R/Fc Chimera (s-IL-21R), the Cytofix/Cytoperm reagent are from R&D Systems (Lille, France).
In contrast, neutralization of IL-10 with specific mAbs or of IL-21 with the recombinant IL-21R/Fc chimera (s-IL-21R) had no effect (data not shown).
Since exposure of the mb-IL-15 present on PB-HP to the soluble IL-15Rα/Fc Chimera (s-IL-15Rα) induces a reverse signal activating the MAPkinase ERK (Fig. 1F), we investigated if other signaling pathways were modified after incubation of PB-NKP with the soluble receptor.
Strong growth arrest paralleled with PFUS1-reporter activity was also observed for the converse receptor chimeras, s-Fz1/2-Ste2(i), which employ the Ste2-intracellular portions, expected to afford good coupling to yeast Gpa1p.
To test which domain(s) of Chd1 are responsible for its measurement activity, we generated chimeras between K. lactis and S. cerevisiae Chd1, and we tested these chimeric proteins for longer measurement in the S. cerevisiae genome.
However, a remaining concern of the S-Mpl chimera may be the magnitude of the cellular expansion level driven by this molecule, which was significantly less than that mediated by endogenous wild-type c-Mpl.
Expectedly, upon overexpression of the s-Fz1/2-Ste2(i) chimeras in a yeast wildtype strain this growth behaviour was paralleled by significant inducible activity over the OFF-strain in the PFUS1-luciferase reporter system, similar to what was observed for native s-Fz1 and the Ste2-Fz1(i) chimera (Fig. 8A).
The far1 effect is particularly obvious in the case of the s-Fz1/2-Ste2(i) chimeras, where complete growth arrest is observed under induced conditions for the wildtype strain.
When we tested the s-Fz1/2-Ste2(i) chimeras in the growth modulation assay, we observed that both receptors of this type exert a complete growth arrest (Fig. 3C), even more pronounced than the α-factor response in wild type (Fig. 3A, right panel).
