The relatively minor change in Km value (from 2.7 to 3.2 mg mL−1) after the immobilization suggested that PEGylated GO-Cellulase was capable of closely mimicking the performance of free enzyme.
However, our kinetic analysis shows a substantial effect on kcat with little change in KM, consistent with results from a similar study with the Thr313Ile mutant of AMSH.
For our triple mutant (B6-III), the change in KM values appeared to be mostly additive, raising the apparent binding constant for d- and l-Thy to 632 and 252 μM, respectively.
For DAS, on the other hand, the effect was more subtle, in which kcat was reduced from the value of AAS by 20% and there was almost no change in KM.
An explanation for the change in Km tolerance in sor1 is that the mutation affects the expression of the NPTII gene which is an integral part of the exon-trap.
The kinetic constants showed a steady reduction in Vmax with increasing concentration of 4′-OH-CB112 4′-OH-CB112 4′-OH-CB112m, indicating a noncompetitive type of inhibution (Tablittle

Changes in Km and Vmax (compared to non-inhibited Rv3802) were determined using Prizm 5.0 software, as were apparent Ki and IC50 values.
Changes in KM upon mutation may arise from changes in the substrate dissociation constant and/or from changes in other microscopic rate constants.
The majority of this effect stemmed from a decrease in kcat values (30−150-fold) rather than changes in KM values (4−6-fold), consistent with an important role for these residues in the lactone hydrolysis mechanism.
The observed changes in Km and kcat for isoleucine and valine activation are quite small for the Y59F variant, demonstrating that the phenolic hydroxyl group of tyrosine does not play a significant role in the amino acid activation step.
The influence of cAMP-dependent kinases on AA-NAT may include two phases related to a different affinity of phosphorylation sites in the enzyme molecule or/and in other regulatory molecules, which are responsible for the stabilization of AA-NAT at the first stage and the changes in Km at the second stage.
