Levy et al. [26] reported the effective use of a randomly mutated BoNT/A heavy chain library, displayed on the surface of yeast cells, to identify single amino acids important in the epitopes of anti-BoNT/A monoclonal antibodies.
5 µg of each VH chain library as well as 1,5 µg of the VL fragments were digested using 50 U NotI and 50 U MulI (NEB) in a 100 µl reaction volume overnight at 37°C.
Five micrograms of each VL chain library, as well 1.5 μg of the VH fragments, were digested using 50 U NcoI and 100 U HindIII (NEB) in a 100 μl reaction volume (overnight at 37°C).
The selected variable antibody domain genes of the shuffled human light chains were cloned as ApaLI and NotI fragments in pHenIX containing a human variable heavy chain library (8 × 10; R. Finnern, unpublished data) and introduced into E. coli TG1.

One of the major drawbacks to this procedure is that L chain libraries have to be created for each distinct H chain, which involves complicated cloning procedures.
Compared with the gDNA heavy chain libraries, the corresponding cDNA heavy chain libraries had significantly higher percentages of clones using IGHV1 and IGHV3 lineages (Fig. 1), and were biased to certain VH1 and VH3 subfamilies, including IGHV1 18, 1 2 and 1 69, and IGHV 3 11, 3 21, 3 23, 3 30, 3 33, 3 49, 3 7 and 3 74 (Fig. 2).
Among the four gDNA heavy chain libraries, NIgH and pt1gH showed a similar pattern of various IGHV lineage usage, whereas pt2gH and pt3gH were significantly different from NIgH and pt1gH in using IGHV1, IGHV2 and IGHV6 lineages (Figs 1 and 2).
All Saints Church is in Early English and Decorated styles; it, too, has a chained library.
Begun in 1130, with many later renovations, the cathedral contains the largest surviving example of a medieval chained library in the world, with some of its books dating to the eighth century, and Mappa Mundi, the only 13th-century illustrated map of the world.
