To do this we initially used MTS24 expression to fluorescence activated cell sort for embryonic (E13.5) thymic epithelial cells from wild-type and K14CreERxRac1flox/flox mice.
Conditions for the cell sort were established first by defining the parameters for pTURBO-gfp (non-fluorescent negative control), blended filtered tomato pulp (negative control), a strongly fluorescent construct carrying promoter for dppA (positive control) and filtered tomato pulp spiked with the dppA-gfp construct (Fig. 7).
Cell sorting was carried out using cell sort precision mode, with a 70 μm nozzle.
Sorting was performed at 2,000 events/s flow rate using 'single cell' sort precision mode, with a 100 μm nozzle.
This is precisely the reason as to why in CD8+ T cells the overlap is higher in the two DNA aliquots or in the same T cell sort of the same individual as compared to the CD4+ T cells.
Of special interest are the results derived from the comparison of the T cell composition of (i) different DNA aliquots from the same DNA preparation of the same T cell sort, (ii) different T cell sorts of the same individual at the same time point and (iii) sorted T cells of different individuals.
