A discrepancy is visible in the case of spherical aberration, for which the dip at position ≈ 100 μm does not appear in the simulations.
MTS provides biased estimates especially in the case of recurrent aberrations.
However, as the BAF approaches 0.5 (as occurs in the case of subclonal aberrations) the two distributions increasingly overlap, resulting in wider confidence intervals in the estimated BAF.
In order to enable three-dimensional (3D) super-resolution imaging, the axial symmetry of the PSF has to be broken, that is, in conventional microscopy (in the case of zero aberrations), the PSF of a fluorophore slightly above and below the image plane appears equally blurred preventing the extraction of the accurate axial fluorophore position.
Several cases of chromosomal aberration encompassing the human chromosome 21q22.3 up to the qter regions have been reported (Lyle et al. 2009; Hannachi et al. 2011; Melis et al. 2011; Roberson et al. 2011) with mild phenotypes.
In the case of spatially-resolved aberration mapping, the P×N acquired images are split into sub-images of size L×L pixels and the same process for aberration retrieval is subsequently used on each sub-image to obtain, for each mode i, a map { a im k, l)} kl that has a lateral resolution of L×p where p is the size of a pixel in the original image (Fig. 1).
For I-FISH, a low tumour cell number can also be sufficient, in case of numeric/structural chromosome aberrations.
Contrary to the case of human skin, aberrations arise here mostly from in-depth heterogeneity in the tissue rather than from the surface of the sample.
Example 3. We simulate e ij's from AR(1) model in Example 2. We consider three cases of large aberrations containing 99.8%, 80%and50%0% of the probes, respectively, in each profile.
It is postulated here that when a growing OF has progressed past the priming region and its main portion has been synthesized, upon approaching its conclusion, the final elongation may take place in a region of the template where certain triplets are repeated: in that case a series of aberrations on the elongation mechanism may occur.
