Standard ChIP analysis showed the presence of H4K16ac at candidate replication origins.
In this study, we show that SMARCA5 associates with nascent DNA, and its occupancy on chromatin and at candidate replication origins increases in S-phase.
Interestingly, we find an inverse correlation for the occupancy of H4K16ac and SMARCA5 at candidate replication origins (α- globin, β- globin and pancreatic amylase) in S-phase cells.
Consequently we propose that the inner region of a tumor confers environmental resistance to cells there residing due to reduced blood flow, hypoxia, and acidosis and that it also harbors most of the phenotypically resistant cells as well, making this area a candidate for origin of tumor regrowth and source of chemo resistance.
No orc/cdc6 genes are encoded by either pNG400 or pNG200, and no candidate replication origin was identified adjacent to the orc/cdc6 gene in pNG300, indicating that other types of replication origins are involved in the initiation of replication in these replicons.
