The pHi dependence of intracellular buffer capacity (βi; i.e. the buffering power of all non-HCO3, non-CO2 buffers) was computed using the NH4 pulse method, and the actual activity of the exchanger in terms of proton flux rate (mmol/l H+/min) was determined by multiplying the rates of pHi change by the cells' intrinsic buffering capacity (βi) at the pHi at which the measurement was taken.
External pH was changed using a mixture of three buffers to minimise changes in buffering power.
Furthermore, the buffer in general has more buffering power in the acidic region (the right side of the chamber) than in basic region (the left side of the chamber) due to the higher starting concentration for HPO4 2– (4 times higher than that of H2PO4 –).
