As shown in Figure 2, both the SQ1A SQ1B and SQ1A C2 duplexes incubated in potassium-containing buffer form species that migrate more slowly in the gel than the 39-mer homoquadruplexes of C2 and SQ1A. Indeed, the inter-conversion of ADP ribosylated and AMPylated BiP to the unmodified active form follows a coherent pattern whereby increased client protein load converts the inactive buffer form to the active one and diminished client protein load pushes modification the other way (Laitusis et al., 1999; Ham et al., 2014; Sanyal et al., 2015).
The oligomers have attenuated client binding capability and may convert to the more active monomer (Freiden et al., 1992; Blond-Elguindi et al., 1993; Chevalier et al., 1998; Thompson et al., 2012; Aprile et al., 2013), suggesting that they may represent a rapidly accessible, inactive buffer form of chaperone.
Since the naphthyl group promotes formation of hydrogels from small peptides, we connected an unnatural amino acid with a naphthyl group (2-Nal) to fMLF to obtain fMLF- 2-Nal) (1). 1 selfMLF- 2-Nal in Dulbecco's Phosphate-BufMLF- 2-Nalne (DPBS) buffer to form a supramolecular hydrogel at a concentration of 0.2 w/v%.
Once in the matrix, calcium is buffered by forming reversible calcium phosphate complexes.
Motif analysis is also consistent with BQS: there is a much higher proportion of unregulated transcription factors than would be expected by chance, and the proportion of stable 3- and 4-node motifs is heavily biased towards the buffered stable forms that enhance BQS.
Along with AE3c, CAII and CAXIV are also involved in tight regulation of horizontal cells pHi, increasing the effectiveness of the bicarbonate buffer, forming metabolon-like systems [26] (Fig. 7B).
The pellet was again resuspended into 1 mL buffer forming the final sample corresponding to the membrane fraction.
