Western Blot analysis independently confirmed the immunohistochemical findings of E-cadherin, GSK3β and Slug proteins expressions in IDCs of breast and paired normal breast tissues (data not shown).
No significant differences in SIRT1, SIRT2 and SIRT3 gene expression were observed in a simple comparison of biopsies from breast cancers with 'normal' breast tissue (data not shown).
The carrier case was diagnosed before age 30 years, was Caucasian, and reported one first-degree relative with breast cancer who was diagnosed after age 45 years and no family history of ovarian cancer, bilateral breast cancer, or male breast cancer (data not shown).
In both the cohort and case control analyses, they found significant positive associations between age at first birth and breast cancer (data not shown) and nulliparity and breast cancer (relative risk (RR)=2.32, 95% CI: 1.45, 3.70 in the cohort analysis; case control data not shown).
Associations of ER+ and ER– breast cancer subtypes with dietary cadmium intake were similar to overall results for breast cancer (data not shown).
These also confirmed MDA-MB-231 breast cancer cells secrete much higher levels of IL-6 than immortalized MCF-10A breast cells (data not shown).
Among current users, there was no association between weight gain and risk of breast cancer; however, this analysis was limited by low statistical power (only 2322 women and 82 cases of breast cancer) (data not shown).
Loss of FBXW7/hCDC4 expression is unlikely to result from allelic imbalances of the 4q31 locus, as examination of >140 primary breast tumors using Representational Oligonucleotide Microarray Analysis (ROMA [ 40]), showed that FBXW7/hCDC4 is hemizygously deleted in less than 20% of primary breast tumors (data not shown, A. Zetterberg and P Lundin, personal communication).
We also found no differences in the levels of the inflammatory cytokine IL-1β between CTR and HFD breast milk (data not shown).
The frequency at which tumor tissues were scored as differentially methylated at these loci was not significantly associated with patient, age, estrogen receptor status or family history of breast cancer (data not shown).
