Human skin fibroblasts (ATCC, #CRL-2703) were used as a negative control, while human patellar tendon stem cells, isolated according to the protocol by a previous study [ 9], were used as a positive control; both cell types received the same treatments as rabbit TSCs.
Both cell types received substantial excitatory input in complete darkness as evinced by the suppression of holding current and noise by 10 µM NBQX, an AMPA receptor antagonist (data not shown); however, spontaneous current fluctuations observed in AII amacrines were much larger in amplitude (σ = 1000 ± 154 pA, n = 20) than those observed in A17 amacrines (σ = 72 ± 22 pA, n = 17; Figure 1E).

A recent study has identified some of the cell types receiving close appositions from PPG axons within the brainstem [ 21 ].
In contrast, the KCs of the MB comprise 2000 neurons that define only seven distinct cell types, receive unstructured, rather than stereotyped, inputs (Murthy et al., 2008; Caron et al., 2013; Gruntman and Turner, 2013), and connect to multiple MBONs through plastic synapses (Cassenaer and Laurent, 2007, 2012).
To parameterize the model, we estimate the number of inputs that a neuron of a given cell type receives from the experimental data.
Animals were divided into 3 groups: group I (n = 6) received a PGA/PLA scaffold saturated with hydrogels not containing cells; group II (n = 6) received scaffolds seeded with both cell types suspended in saline rather than hydrogels; and group III (n = 6) received scaffolds seeded with both cell types suspended in hydrogel composites.
Animals passively transferred with both cell types were fully protected against morbidity (Figure 7A) and mortality (Figure 7B), while animals that received only one cell type respond poorly to infection, with a significant weight loss between days 10 and 15 p.i. (Figure 7A).
Both cell types displayed similar viscoelastic properties.
However, if one were to perform a traditional gene expression analysis comparing c-Myc cells with cells without c-Myc, one would "control" for these differences in RNA amount by using the same amount of RNA for both cell types.
