Out of 34 cell lines for which transcriptomic data were available (from our own dataset: 25 cell lines upper panel, or from publicly available data: 22 cell lines lower panel, 13 cell lines in common; Fig 7A), we identified two with strong expression of the DCN mRNA, TCCSUP and HS172T.
Of 616 miRNAs, 75 showed a >2-fold increase in the Mim23 cell line relative to both the WT- and Mut23-expressing cell lines (upper-right quadrant of Figure 5B; Tables S4 S6).
Although at this very low concentration of paclitaxel we detected no cell death in either cell line (white bar in Figure 2G, upper), addition of 1 μM additively increased the effect of IRIS peptide on the survival of both cell lines (dark-colored lines in Figure 2G, upper).
As shown above, survival gradually decreased with increasing IRIS peptide concentration in both cell lines (light-colored lines in Figure 2G, upper), which correlated very well with gradual increase in activated caspase 3 and 7 in both cell lines.
Efficacy of RH1 in SRB assay correlated well with efficacy of cisplatin, and within tumour type the cell line with the higher DTD activity was the more sensitive to RH1, even when, as with the osteosarcoma cell line pair, both cell lines had DTD activity above the postulated upper threshold.
Measles virus infection triggers the IFN response in human Huh7 cells, but not in the RIG-I defective Huh7.5 cells, although the accumulation of N mRNA reached a similar level in both cell lines after 24 h (Figure 3A lower and upper panel, respectively).
Thus, while real-time quantitative PCR (qRT-PCR) measurements showed that the replication of R. raoultii was similar in both cell lines, (Fig. 5) the L929 monolayer was not at all damaged by R. raoultii (Fig. 6, upper left panel).
Both cell lines were incubated with Sgc8-FSNPs.
