MO and HSL (as part of their PhD studies) carried out the experiments on CD47/IAP expression by immunohistochemistry, co-immunoprecipitation, and Western blotting in addition to undertaking analysis of signalling events and changes in cell membrane potential following mechanical stimulation of chondrocytes.
Some of the squamous cell carcinomas were tested by Southern blot hybridization in addition to PCR.
The reduction of actin expression was detected by immunohistochemistry and western blot techniques, in addition to severe damage in the morphology of tegument [ 95].
We examined mtDNA from MGME1-null patient fibroblasts by Southern blotting, and in addition to the previously reported reduction in mtDNA copy number, we also observed an mtDNA fragment below the expected restriction size (sub-genomic fragment) (Fig. 3A).
Western blots confirmed that in addition to MITF, protein levels of MITF transcriptional targets CDK2, BCL2, and MLANA were also markedly decreased in lower MITF Motif 1 cell lines (NZM09, NZM11, NZM40) compared to Motif 2 (Figure 1C).
As listed in Table S4, 11 of the 12 spots have been reported to exist in the apoplast (except for spot 40), and most of the 11 spots have been detected in the apoplast by Western blot or immunocytochemistry in addition to proteomics [12], [27], [28], [34] [40], with only two spots (spots 45 and 51) detected just by proteomic approaches [27], [41].
In mitochondria isolated from cultured fibroblasts complex I activity was almost undetectable in patient 1, and 50% reduced in patient 2. In fibroblasts Southern blot analysis identified, in addition to the wild type molecules, two smaller bands sized approximately 11.0 Kb in patient 1 (Fig. 2A) and 13.0 Kb in patient 2 (Fig. 2B).
The majority of lanes in the Western blots had at least one additional band in addition to the band running at the predicted molecular weight, although these other bands were usually lighter than the band at the predicted molecular weight.
