CD28 blockade of Th17 development was correlated with a decrease rather than an increase in the percentage of Foxp3+ T cells.
Intriguingly, the neutralization of one of these two cytokines (IL-10 or TGF-β) just had minor effects on Treg-mediated blockade of diabetes development (data not shown), indicating a complementary role of IL-10 or TGF-β in mediating Treg protective effects when one cytokine was removed.
In this case, TNF-alpha-mediated DKK1 represents represents a major factor in the blockade of osteoblast development, resulting in the inflamed joint going unrepaired [ 15].
In contrast, downregulation was observed in several processes that indicate the blockade of active development: microtubular transport, spermatogenesis, mRNA processing, and the enzymatic complex of mitochondrial TCA cycle (Table 2).
These changes probably resulted in blockade of direct development and deep restructuring of metabolic pathways indicated by differential expression of genes involved in cell cycle regulation, metabolism, detoxification, redox balance, protection against oxidative stress, cuticle formation and synthesis of larval storage proteins.
Importantly, in the presence of IL-6, TGF-β induced development of Treg cells was blocked, whereas blockade of IL-6 permitted development of Foxp3+ Treg cells, suggesting that IL-6 inhibited Treg development while enhancing Th17 development induced by TGF-β [ 17, 24, 25].
However, the effects of Dll4 inhibition on angiogenesis are clearly heterogeneous, at least in normal tissues, as evidenced by the lack of appreciable effect of Dll4 blockade on follicular development.
However, the lack of an efficient means to test immune cell responses to the abundant combinations available represents a significant blockade on the development of new vaccines.
We also found that additional HER2 blockade after the development of lapatinib resistance was ineffective and that PI3K inhibition was essential to overcoming resistance.
Strategies that are in various stages of development include blockade of the extracellular receptor domain [ 44] and inhibition of the intracellular tyrosine kinase activity [ 53].
The time-course of development of blockade was well-described by a mono-exponential fit to the data, with a rate constant (K) for the fit of 16.33 s−1 (equivalent to a time-constant (1/ K) of 61 ms).
