The role of the oncosuppressor p53 in the regulation of autophagy in cancer cells appears ambiguous: while nuclear DNA-binding proficient p53 promotes the transcription of certain autophagy genes [ 51], p53 mutants that reside in the cytoplasm hamper autophagy [ 52, 53].
There are approximately 200 DNA binding-proficient Atf1 protein molecules per cell [11] and there are about 300 to 800 DNA binding sites for Atf1 per genome (depending upon whether one uses a strict or relaxed consensus for the CRE-like, M26 DNA site) [13], [16].
In control experiments, we showed that cysteine accessibility of the binding-proficient Ste2-Y106C-HA receptor was sensitive to α-factor, whereas cysteine accessibility of the Ste2-Y106C, F204S-HA was not sensitive to α-factor when it was expressed alone (Fig. 5A).
Researchers have focused the production of non-biodegradable, antibiotic, infused bone cements specifically designed for proficient osteocyte binding, useful antibiotic release over a desirable period of time, and promotion of bone regeneration.
As shown in Figure 7A, immunoprecipitation assays revealed that both membrane-bound CD44-CP1 and soluble Flag-CP1 were proficient in binding TSC2.
Thus, all of the residues in human FKBP12 required to interact with Tor are sufficiently conserved indicating that the human FKBP12-rapamycin complex is fully proficient in binding and inhibiting the yeast Tor1 and Tor2 proteins.
We thus conclude that the mutants all show proficient DNA binding.
Cas8′ mutant proteins were proficient in binding to R-loop +PAM (Supplementary Figure S5) and other substrates (result not shown) and interacted with MBP Cas5-Cas7 (Supplementary Figure S6).
FtsZD210G, on the other hand, is proficient in GTP binding, but is defective in GTP hydrolysis [28].
Of these four, only the M362L variant was proficient at heme binding: a gel filtration step intended to purify hemoprotein after heme reconstitution resulted in mainly apoprotein in the cases of the Y440A, Y444A, and S361A variants (data not shown).
This held true even if the replacing non-cohesive FG domain was fully proficient in NTR-binding.
